This comprehensive encyclopedic reference provides rapid access to focused information on topics of cancer research for clinicians, research scientists and advanced students. Given the overwhelming success of the first edition, which appeared in 2001, and fast development in the different fields of cancer research, it has been decided to publish a second fully revised and expanded edition. With an A-Z format of over 7,000 entries, more than 1,000 contributing authors provide a complete reference to cancer. The merging of different basic and clinical scientific disciplines towards the common goal of fighting cancer makes such a comprehensive reference source all the more timely.

Angiogenesis is the process through which new blood vessels develop from pre-existing ones. It is an important process of growth and development and aids in wound healing and formation of granulation tissue. Angiogenesis may be stimulated mechanically and chemically. Though mechanical stimulation is not well characterized, chemical stimulation is done using angiogenic proteins such as integrins and prostaglandins, and growth factors like vascular endothelial growth factor (VEGF), fibroblast growth factor (FGF), etc. The application of angiogenesis in medicine is as a therapeutic target for the management of abnormal vasculature or poor vascularization and cardiovascular disease. This book brings forth some of the most innovative concepts and elucidates the unexplored aspects of angiogenesis. It aims to give a general view of the physiology and pathology of angiogenesis. The extensive content of this book provides the readers with a thorough understanding of the subject.

Angiogenesis describes the formation of new blood vessels, which arise as outgrowths from existing vessels. In many physiological processes such as ovulation and wound healing angiogenesis is involved for a relatively short time. Otherwise under normal physiological conditions in the adult organism angiogenesis is an extremely slow process. By contrast in certain disease states such as diabetic retinopathy, arthritis, chronic inflammation, hemangiomas, etc., angiogenesis persists and contributes to the pathology of these disease states. Some 50 such "angiogenic diseases" have been described where angiogeneSiS is involved. Also in tumor growth and metastasis angiogenesis is an essential process and precedes neoplastic transformation. Hence, angiogenesis could become an important diagnostic tool and a target for developing therapeutic agents. This book contains the proceedings of the NATO Advanced Study Institute on "Angiogenesis in Health and Disease" held in Porto Hydra, Greece, from June 16-27, 1991. This meeting was a comprehensive review of endothelial cell biology and endothelial cell phenotypic and functional heterogeneity in relation to angiogenesis under physiological and pathological conditions. Numerous in vitro and in vivo models were presented, which are used to study angiogenesis at the molecular and cellular levels and to evaluate chemical compounds or naturally occuring substances for their effect on angiogenesis. The presentations and discussions at this meeting provided an opportunity for the basic science and the clinical disciplines to meet, exchange information and provide future research directions for many investigators engaged in the study of angiogenesis.

Angiogenesis is a highly complex phenomenon where new blood vessels are formed for the supply of oxygen and nutrients in different organs of the body. It plays a critical role in both physiological processes such as growth and development as well as pathological processes including cancer and different types of tumors. Angiogenesis is also essential for the regeneration and survival of cells in several disease conditions such as ischemic heart disease (myocardial infarction), atherosclerosis, brain injury (stroke) and diabetes. Since the mechanisms of angiogenesis are organ specific and differ among various diseases, it is proposed to devote one section of this book to the development of angiogenesis in some selected diseases such as cancer, ischemic heart disease, atherosclerosis, diabetes and stroke. It is pointed out that extensive research work in this regard has been carried out in the area of cancer and heart disease, whereas relatively less attention has been paid to studying angiogenesis in other disease conditions.

An international team of experts critically review the recent progress in basic and applied research in angiogenesis. Their cutting-edge discussion ranges from the stimulation and repression of angiogenesis to the discovery of novel targets and the use angiotherapy in the clinic. They also detail the fundamental concepts in the physiology and pathophysiology of angiogenesis and evaluate the potential of angiotherapy in the management of angiogenic disease, highlighting some of the angiogenics and antiangiogenics both in development and in clinical trials.

The microcirculation is highly responsive to, and a vital participant in, the inflammatory response. All segments of the microvasculature (arterioles, capillaries, and venules) exhibit characteristic phenotypic changes during inflammation that appear to be directed toward enhancing the delivery of inflammatory cells to the injured/infected tissue, isolating the region from healthy tissue and the systemic circulation, and setting the stage for tissue repair and regeneration. The best characterized responses of the microcirculation to inflammation include impaired vasomotor function, reduced capillary perfusion, adhesion of leukocytes and platelets, activation of the coagulation cascade, and enhanced thrombosis, increased vascular permeability, and an increase in the rate of proliferation of blood and lymphatic vessels. A variety of cells that normally circulate in blood (leukocytes, platelets) or reside within the vessel wall (endothelial cells, pericytes) or in the perivascular space (mast cells, macrophages) are activated in response to inflammation. The activation products and chemical mediators released from these cells act through different well-characterized signaling pathways to induce the phenotypic changes in microvessel function that accompany inflammation. Drugs that target a specific microvascular response to inflammation, such as leukocyte-endothelial cell adhesion or angiogenesis, have shown promise in both the preclinical and clinical studies of inflammatory disease. Future research efforts in this area will likely identify new avenues for therapeutic intervention in inflammation.
