Cellulase is a key enzyme of industrial interest and plays a crucial role in the hydrolysis of cellulose, a prime component of plant cell walls. Cellulase covers a broad area in the global market of industrially important enzymes and it is considered as the third largest industrial enzyme globally. Additionally, cellulase contributes about 20% of the total enzyme market globally because of its massive demand in various industries such as in biofuel production, pulp, paper, textile, food, and beverages, as well as in detergent industries. Among these, the demand of cellulase may become frequently selected in the commercial production of biofuels in the future and thus will further increase demand of cellulase in the biofuel industry. Because biofuel production is still not realized in a cost-effective, practical implementation due to its high cost (the higher cost of biofuels is due to higher production costs of enzymes), there is a need to introduce these types of approaches, which will help to lower the cost of enzyme production for developing overall economic biofuel production.

Deswegen ist seine genauere Erforschung in der Bioethanol Produktion von großer Bedeutung. Ein wichtiger Aspekt dieser Forschung beruht auf der industriellen Stammverbesserung von nur einem einzigen Isolat, dem Stamm QM6a. Die Entdeckung der Möglichkeit von T. reesei zur sexuellen Fortpflanzung, eröffnete neue Wege in der Forschung hinsichtlich der industriellen Stammverbesserung abseits gentechnischer Methoden (Schmoll, 2013 Seidl et al., 2009 Schuster and Schmoll, 2010).Im Rahmen dieser Masterarbeit wurden die Paarungstypen von isolierten Trichoderma spp. aus dem Iran und Neuseeland analysiert, um bereits etablierte Bedingungen für die sexuelle Entwicklung und Fortpflanzung von T. reesei (Seidl et al., 2009) weiter zu optimieren. Die daraus gewonnenen Erkenntnisse sollen dazu genutzt werden, auch in anderen, industriell interessanten Trichoderma Stämmen die sexuelle Fortpflanzung unter Laborbedingungen zu ermöglichen. Zusätzlich wurden verschiedene Trichoderma Stämme aus österreichischer und deutscher Erde isoliert, um die eigene Stammsammlung zu erweitern und weitere Kreuzungen durchzuführen, um so Stammverbesserungen ohne Einsatz gentechnischer Methoden zu ermöglichen.*****The fungus Trichoderma reesei, which is one of the most prolific cellulase producers, is considered as an ideal model system to study the regulation of plant cell wall degrading enzymes. Those enzymes are required for production of second generation biofuels. Nevertheless, this industrial application is not yet economically sustainable. After the finding that T. reesei can undergo a sexual reproduction, a new tool for research and industrial strain improvement was established (Schmoll, 2013 Seidl et al., 2009 Schuster and Schmoll, 2010). This reason has increased the interest of the industry in the improvement of the strains used for production of cellulases.In this study, mating types of available Trichoderma spp. were analyzed to establish further investigation of sexual development in other species than T. reesei, whose sexual reproduction was already confirmed. In addition, several Trichoderma spp. from Austrian and German soils were isolated to extend our own strain collection and enable further crossing approaches, which will help to achieve strain improvement without genetically modified techniques. Another part of this master thesis focused on the potential possibilities for improving the crossing conditions, which covered a broad evaluation of environmental factors and nutrient types. These results could help to improve the sexual development in T. reesei under laboratory conditions and also serve as reference for sexual development in other Trichoderma species.
