DOWNLOAD EBOOKCalvaria dissected from 6-day-old mice were maintained in organ culture in a chemically-defined medium without serum additives. Bone resorption was quantified by the release of 45Ca or hydroxyproline (Hyp) into the medium. Collagen synthesis (Hyp) in bone was estimated by the incorporation of [5-3H]-proline. Prostaglandin E2 (PGE2), prostaglandin F (PGF) and 6-keto-prostaglandin F1a (6-keto-PGF1a) released into the medium from cultured calvaria, were measured by radioimmunoassay. Prostanoids were also quantified by the release of radioactively-labelled metabolites from calvaria pre-labelled in vitro with [3H]-arachidonic acid. These metabolites were separated by high performance liquid chromatography.
