This volume represents the proceedings of the Fifth Congress of the International Society of Ocular Toxicology (ISOT), which was held at the Grove Park Inn and Resort in Asheville, North Carolina, October 13-17, 1996. We are delighted to present this volume to the ophthalmic community, especially those with a significant interest in ocular toxicol ogy. The Fifth Congress was developed around themes relating to ocular drug metabolism, the ocular pathophysiological effects of nitric oxide, government issues relating to the use of alternative methods for toxicity testing, and a workshop that encompassed comparisons of both in vitro versus in vivo models as well as different animal models. The outcome of this congress, embodied in this volume, is a contribution to the methodologies currently employed or under development and to various drug or physical effects on different ocular tissues. While the focus of this proceedings is on ocular effects of drugs or other materials, many of the contributions deal with topics that have a much broader interest. The workshop concerning the use of different model systems and the choice of the best animal model for drug testing covers a wide range of interests that ex tends far beyond specific ocular effects. This is especially true in the area of alternative methods and in the choice of the best animal model for examination of different disease entities.

This volume presents the latest findings regarding the control of the fluid environments of the eye and brain. The book is comprised of papers presented at a symposium honoring the 80th birthday of Professor Hugh Davson. The contributors are previous students of Hugh Davson who are now leading authorities in the fields of the eye and brain. The eye is considered from its many interfaces, including the role of prostaglandins, blood flow in diabetes, and the vitreous body. Other topics discussed include the role of carbonic anhydrase, cerebrospinal fluid and its drainage, and the development of the blood/brain barrier to macromolecules. The book also shows how recent findings about homologous molecules refute earlier studies with foreign tracers, and offers a fascinating glimpse into the future with a description of the latest imaging techniques used in neurological diagnosis.

A significant new book on drug delivery to the eye Biopharmaceutics of Ocular Drug Delivery examines the current physiological, anatomical, pharmaceutical/technological, and biopharmaceutical knowledge on ocular drug delivery. Leading ophthalmic researchers cover topics such as the formulation and usage of ophthalmic solutions and suspensions, the influence of surfactants and preservatives on ocular drug absorption, and new techniques that include bioadhesion and the use of hyaluronic acid as an ophthalmic vehicle. Promising research on new delivery systems for improved drug therapy is highlighted, including new pharmaceutical vehicles, inserts, liposomes, nanoparticles, and prodrugs. Biopharmaceutics of Ocular Drug Delivery is one of the few books available on drug delivery to the eye and will be an important reference for ophthalmologists, pharmacologists, pharmaceutical researchers, and other scientists interested in the topic.

This volume presents a basic consensus of how the aqueous humor is formed and exits through the trabecular meshwork and canal of Schlemm. It presents a timely update to current knowledge of the molecular transport mechanisms which underlie aqueous humor dynamics. In addition, it provides a concise description of the clinical approaches used for assessing these basic transport processes. The book emphasizes the phenomenon of the diurnal rhythm of aqueous humor formation, from both the clinical and molecular points of view. This phenomenon provides the major indication that aqueous humor secretion is regulated. - Introduces the mechanisms of aqueous humor formation and outflow - Describes the clinical study of aqueous humor dynamics - Exposes the research approaches currently applied - Emphasizes the diurnal rhythm of aqueous humor secretion - Provides awareness of unanswered questions

The purpose of this book is to present a synopsis of what is known about substances that have toxic properties injurious to the eyes, disturbing to vision, or affecting eyes in other unwanted ways. The coverage is truly comprehensive, encompassing local and systemic, acute and chronic, human and veterinary toxicology of the eye. The text summarizes mechanisms of injury, treatments, and other relevant knowledge for more than 3000 alphabetized substances - essentially all those on which public information is available. Also described are systemic side effects of ophthalmologic drugs, treatment of chemical burns of the eyes, and testing methods and species specificity for toxic effects on the eyes. Facilitating access to this prodigious amount of information is a large index that cross-references substances and effects, including numerous synonyms. This monumental work is a truly definitive text and a highly useful reference book that should be available to every ophthalmologist, emergency room, and medical library.

Ocular toxicity is routinely assessed in toxicology studies conducted for regulatory purposes. Ocular anatomy and physiology and the assessment of ocular toxicity itself can be challenging to scientists involved in the safety assessment of pharmaceuticals, pesticides and other agents. Anatomical and physiological differences between species can impact the nature of ocular effects observed following intended or unintended exposure of ocular tissues to xenobiotics. Ocular Toxicity in Laboratory Animals provides a concise reference addressing ocular anatomy and physiology across species that will enhance the design and interpretation of toxicology studies conducted for regulatory purposes. The book provides an overview of routine and advanced techniques that are used to assess ocular toxicity including slit lamp biomicroscopy, indirect ophthalmoscopy, electrophysiology and imaging methods for the anterior and posterior segments of the eye. Additionally, the book defines the regulatory expectations for pharmaceuticals intended to treat ocular diseases and for other non-pharmaceutical regulated chemicals. With contributions from experts in the field, Ocular Toxicity in Laboratory Animals is an authoritative, accessible guide for toxicologists and other scientists involved in conducting toxicology studies for regulatory purposes and/or reviewing data from such studies.
