Bone resorption was quantified by the release of 45Ca or hydroxyproline (Hyp) into the medium. Collagen synthesis (Hyp) in bone was estimated by the incorporation of [5-3H]-proline. Prostaglandin E2 (PGE2), prostaglandin F (PGF) and 6-keto-prostaglandin F1a (6-keto-PGF1a) released into the medium from cultured calvaria, were measured by radioimmunoassay. Prostanoids were also quantified by the release of radioactively-labelled metabolites from calvaria pre-labelled in vitro with [3H]-arachidonic acid. These metabolites were separated by high performance liquid chromatography.
