Time-resolved fluorescence spectroscopy is widely used as a research tool in bioch- istry and biophysics. These uses of fluorescence have resulted in extensive knowledge of the structure and dynamics of biological macromolecules. This information has been gained by studies of phenomena that affect the excited state, such as the local environment, quenching processes, and energy transfer. Topics in Fluorescence Spectroscopy, Volume 4: Probe Design and Chemical Sensing reflects a new trend, which is the use of time-resolved fluorescence in analytical and clinical chemistry. These emerging applications of time-resolved fluorescence are the result of continued advances in laser detector and computer technology. For instance, pho- multiplier tubes (PMT) were previously bulky devices. Miniature PMTs are now available, and the performance of simpler detectors is continually improving. There is also considerable effort to develop fluorophores that can be excited with the red/ne- infrared (NIR) output of laser diodes. Using such probes, one can readily imagine small time-resolved fluorometers, even hand-held devices, being used fordoctor’s office or home health care.

As stated by its first editor, Dr. D. B. Roodyn, the primary goal of the series Subcellular Biochemistry is to achieve an integrated view of the cell by bringing together results from a wide range of different techniques and disciplines. This volume deals with the applications of fluorescence spectroscopy to membrane research. It seeks to present complementary biochemical and bio physical data on both the structure and the dynamics of biological membranes. Biophysics and biochemistry are improving more and more in their ability to study biomembranes, overlapping somewhat in this area and explaining the functioning of the whole cell in terms of the properties of its individual com ponents. Therefore, we have brought together an international group of experts in order to report on and review advances in fluorescence studies on biological membranes, thereby highlighting subcellular aspects. The first chapters present a critical evaluation of the current applications of dynamic and steady-state fluorescence techniques. Subsequent chapters dis cuss more specific applications in cells, biological membranes, and their con stituents (lipids, proteins).

This volume describes the application of fluorescence spectroscopy in polymer research. The first chapters outline the basic principles of the conformational and dynamic behavior of polymers and review the problems of polymer self-assembly. Subsequent chapters introduce the theoretical principles of advanced fluorescence methods and typical examples of their application in polymer science. The book closes with several reviews of various fluorescence applications for studying specific aspects of polymer-solution behavior. It is a useful resource for polymer scientists and experts in fluorescence spectroscopy alike, facilitating their communication and cooperation.

At the time that the editors conceived the idea of trying to organize the meeting on which the contents of this volume are based and which became, in March 1980, a NATO Advanced Study Institute, the techniques of time-resolved fluorescence spectroscopy, in both the nanosecond and sub-nanosecond time-domains, might reasonably have been said to be coming of age, both in their execution and in the analysis and interpretation of the results obtained. These techniques, then as now, comprised mainly a number of pulse methods using laser, flash-lamp or, most recently, synchrotron radiation. In addition, significant developments in the more classical phase approach had also rendered that method popular, utilizing either modulation of an otherwise continuous source or, again recently, the ultra-rapid pulse rate attainable with a synchrotron source. In general terms, time-resolved fluorescence studies are capable, under appropriate conditions, of supplying direct kinetic information on both photophysics and various aspects of molecular, macromolecular and supramolecular structure and dynamics. The nanosecond and sub-nanosecond time-scales directly probed render these techniques particularly appropriate in studying relaxation and fluctuation processes in macromolecules, particularly biopolymers (e. g. proteins, nucleic acids), in supramolecular assemblies such as cell membranes, and in a variety of relatively simpler model systems.
