The Proceedings of the Second International Symposium on Biochemistry of Exercise are centered on the effects of long lasting exercise and training. In the years following the first symposium which was held in Brussels in 1968, biochem istry of exercise has gained more importance in view of the increasing number of labora tories and scientific papers which are dealing with this field. From the topic of the first symposium - humoral modifications occurring during physical activity - our points of interest have been turned to a more limited area, namely long term exercise and training, It was important to investigate these subjects because everyone knows that in to-day's sport a good performance means hours of weekly or even daily training. Therefore, it was of considerable interest to stimulate discussions and to clarify ideas in this particular field of human activity. Our knowledge of biochemistry of exercise at the cellular level has highly progressed during the last five years. Researchers have focused their interests on the sequential uti lization of fuels, the adaptative responses of the enzyme machinery, the different types of muscle fibers. The topics of the Proceedings include: general subjects, metabolism of carbohydrates, lipids, and proteins, hormonal regulations, electrolytes, ultrastructure and fiber types of muscle, cellular enzymes. The measurement oflactate is straightforward and reliable. The sampling is normal ly dependable, although discrepancies between sample sites can occur if samples are drawn during other than steady state conditions or when blood flow to the site is low. Comparison of data between subjects should consider intra- and interindividu al variation. Interpretation of the significance of blood or even a muscle lactate concentration is extremely difficult and neither can be assumed to reflect rates of lactate release or production. Lactate metabolism appears to be very significant in short-term, high intensity work lasting a few seconds, but the exact role remains to be established. Blood lactate and the pulmonary aspects of anaerobic threshold are not necessarily causally related and the significance of anaerobic threshold as a metabolic measure remains obscure. There are numerous examples of changes in blood or muscle lac tate independent of oxygen availability, and factors such as H+ concentration ap pear to play an important, although undefined, role in regulating lactate production and/or release.

The term cytoskeleton in muscle and nonmuscle cells refers to fibrous proteins that are directly or indirectly associated with cell shape, contractility, and/or motility. These cytoskeletal elements include microtubules and associated proteins, microfilaments and their binding proteins, and lastly, intermediate filaments. Even though the structure and function of myosin and tro pomyosin have been extensively investigated in muscle ceIls, considerably less is known about their functions in nonmuscle ceIls; they are often referred to generically as actin-binding proteins, as are a-actinin, filamin, vinculin, and others. Indeed, most nonmuscle researchers are unaware that the term intermedi ate filament was derived from the observation that the diameter of intermedi ate filaments is intermediate between those of actin (5 nm) and myosin (15 nm) and not intermediate between those of actin (5 nm) and microtubules (25 nm). It has therefore been the purpose of this series to attempt to bridge the communication gap that appears to exist between muscle and nonmuscle researchers. For those students and active scientists who are faced with the task of absorbing new data and concepts at an ever-accelerating rate, review essays are an excellent way to keep up. The essays in this volume, as weIl as in the previous ones, focus on topics of current interest. They are intended to be critical rather than exhaustive.
