The papers in this book were presented at the 14th Mechanisms in B-cell Neoplasia meeting that was held in Bethesda, Maryland October 21-23, 1996. In 1995 the organizers decided that the format of the meeting would be changed and that specific topics relevant to B-cell neoplasia would be discussed. This year's topic is on the c-myc oncogene in B-cell neoplasia which has been discussed in virtually every previous meeting. Some of the presentations announced for the first time dramatic advances in our understanding of c-myc and because this subject has become highly complex it was thought that devoting the whole meeting to this theme would be appropriate. The book, therefore, repre sents a review of many aspects of the myc problem but by no means is truly comprehensive. In a recent Medline search there were 8,505 references to myc, fully illustrating the magnitude of the interest and depth of this field. The organizers of the meeting have each contributed review chapters that summarize different aspects of the meeting. We thank the National Cancer Institute for sponsoring this workshop and the staff of Cygnus, Inc. , for their outstanding organizational assistance. The organizers are most grateful to Vickie Rogers for assembling the book and dealing with the edi torialization of the manuscripts. MICHAEL POlTER FRITZ MELCHERS Table of Contents M. POlTER and K. Genetic alterations in cancer, in addition to being the fundamental drivers of tumorigenesis, can give rise to a variety of metabolic adaptations that allow cancer cells to survive and proliferate in diverse tumor microenvironments. This metabolic flexibility is different from normal cellular metabolic processes and leads to heterogeneity in cancer metabolism within the same cancer type or even within the same tumor. In this book, we delve into the complexity and diversity of cancer metabolism, and highlight how understanding the heterogeneity of cancer metabolism is fundamental to the development of effective metabolism-based therapeutic strategies. Deciphering how cancer cells utilize various nutrient resources will enable clinicians and researchers to pair specific chemotherapeutic agents with patients who are most likely to respond with positive outcomes, allowing for more cost-effective and personalized cancer therapeutic strategies.

The MYC gene family plays essential roles in normal development and in multiple cellular functions. Moreover aberrant MYC gene activation is profoundly involved in the etiology of a wide range of cancers. MYC encodes a transcriptional regulator that modulates expression of genes controlling cell growth, proliferation, metabolism, differentiation, and death. Deregulation of these expression programs has been linked to MYC's function in tumor initiation, progression, and survival. Written and edited by experts in the field, this collection from Cold Spring Harbor Perspectives in Medicine covers all aspects of MYC biology. The contributors discuss its normal functions in the control of cell growth, cell competition, pluripotency, and development, as well as the molecular basis for the effects of the MYC protein on transcription. In addition, they examine how MYC interacts with other proteins, induces apoptosis, and impacts metabolism, genomic stability, and microRNA expression. The authors also provide a detailed analysis of the role of MYC in tumor initiation and progression. Its involvement in cancers such as medulloblastoma, neuroblastoma, and Burkitt's lymphoma is examined, as are the prospects for anti-MYC therapies in cancer treatment. This book is essential reading for all cancer biologists, as well as researchers studying the regulation of gene expression.

This book provides a step-by-step and practically applicable approach for the accurate and clinically relevant diagnosis of lymph node (LN) and bone marrow (BM) biopsies. Clinicians expect pathological guidance not only with accurate diagnosis, but also about disease progression, minimal residual disease, disease susceptibility to a particular therapy, effects of prior therapy on prognosis and subsequent therapy etc. This book provides brief but to the point guidance about the prognostic and therapeutic implications of key ancillary studies so that the pathologist is comfortable to answer clinician’s questions over the entire arc of manifestations and management of the disease. The text follows the WHO (2016) classification in essence but the material is organized in a fashion most useful to a practicing surgical pathologist. This is achieved by focusing on the morphological findings as the starting point. Using this morphological “backbone” and several frequently asked questions (FAQs) the reader is guided to a rational list of differential diagnoses leading to a definitive diagnosis. The contents of each chapter are carefully selected so that the practically important and directly applicable information is available in an easy-to-find and easy-to-grasp format. Practical Lymph Node and Bone Marrow Pathology serves as a practical introduction and handbook for pathology trainees and hematopathology fellows and will remain a useful reference to practicing pathologists when they are signing out lymph nodes or bone marrow specimens.
